Gene linked to male infertility identified

Washington, Sept 16 (ANI): Scientists from Virginia Commonwealth University have identified a gene that may contribute to male infertility.

The research team hopes that the new findings would lead to new approaches to male contraception.

Sperm are produced in the testicles through a three-step process called spermatogenesis.

During the final stage, known as spermiogenesis, a lot of changes take place, including the packaging of DNA into the sperm head and the formation of the sperm tail, which propels the sperm cell toward the egg.

The study conducted using mouse model showed that mice lacking a protein called meiosis expressed gene 1, or MEIG1, were sterile as a result of impaired spermiogenesis – the process that encompasses changes in the sperm head and the formation of the tail.

The team also found that MEIG1 associates with the Parkin co-regulated gene protein, or PACRG protein, and that testicular PACRG protein is reduced in MEIG1-deficient mice.

PACRG is thought to play a key role in assembly of the sperm tail, and the reproductive phenotype of PACRG -deficient mice mirrors that of the MEIG1-mutant mice.

“We discovered that MEIG1 is essential for male fertility. Moreover, our findings reveal a critical role for the MEIG1/PACRG partnership in the function of a structure that is unique to sperm, the manchette. The absence of a normal manchette in mice lacking MEIG1 totally disrupts the maturation process of sperm,” said Dr Jerome F. Strauss III, dean in the VCU School of Medicine.

“In addition to having an impact on fertility, the discovery identifies a new target for drug discovery for a much needed reversible male method of contraception,” he added.

The study is published in the Early Edition of the Proceedings of the National Academy of Sciences. (ANI)

South Africa Sports Minister warns of World War III over Semanya

London, Sep 12 (ANI): The South African government has threatened a “third world war” if the International Association of Athletics Federation bans champion runner Caster Semenya over a test that shows her to be a hermaphrodite, a person with both female and male sexual characteristics.

The IAAF commissioned a gender test on the teenager after her performance levels improved remarkably in the build-up to the competition.

The leaked report that a gender verification test had found the 18 year-old to be a hermaphrodite with no womb was greeted with outrage in South Africa on Friday, with politicians, sports officials and Semenya’s relatives decrying the teenager’s public humiliation.

On Friday the IAAF attempted to diffuse the controversy by refusing to confirm the medical reports, insisting that it would make official comment until after the test results had been verified by a panel of scientific experts.

But the prospect of the teenager being disqualified from future female races drew a furious response from South African Sports Minister Makhenkesi Stofile, The Telegraph reports.

“I think it would be the third world war. We will go to the highest levels in contesting such a decision. She’s a woman, she remains our heroine. We must protect her,” he said.

Semenya was due to run in a 4km race at the national cross-country championships in Pretoria on Saturday, but was withdrawn by her coach Michael Seme on the grounds that she was “not feeling well”.

The IAAF is expected to disqualify the South African from future events and advise her to have surgery because her condition carries grave health risks, The Daily Telegraph report claims.

A source closely involved with the IAAF tests said Semenya had internal testes-the male sexual organs, which produce testosterone.

“There certainly is evidence Semenya is a hermaphrodite. But the trouble is the IAAF now has the whole ANC and the whole of South Africa on their backs. Everything is going to have to be done absolutely by the book, no question of a challenge to the findings,” the source said. (ANI)

DNA analysis key for solving mystery of King Tut’s origins

London, August 26 (ANI): Dr. Zahi Hawass, Secretary General of the Supreme Council of Antiquities, Egypt, has said that harvesting DNA from ancient mummies would be the key to solving the mystery surrounding Egyptian pharaoh Tutankhamen’s origins.

According to an article authored by Dr Hawass in the Asharq Alawsat Newspaper, he was in for a surprise when he entered the royal tomb of Tutankhamen with Professor Zakaria, and managed to get DNA samples.

Previously, there was hardly any hope in obtaining DNA samples from mummies, and Dr Hawass believed that he would prove to the world that mummies did not have any DNA suitable for analysis.

“For the first time, I saw that it is possible to harvest DNA from a mummy, and I believe that this will be the key to solving the mystery surrounding King Tutankhamen’s origins,” he wrote in the article.

Tutankhamen, the golden pharaoh, continues to bedazzle the entire world.

The discovery of King Tut’s tomb, which took place around 85 years ago, remains the most important archeological discovery of our time, not just in Egypt, but in the entire world.

This was the first time that a royal tomb of one of Egypt’s pharaohs was discovered untouched, and with the complete set of funeral furniture which was buried with the King.

In addition to this, there was also the treasures and jewelry which blinded anybody who set eyes on them.

“In an attempt to unravel the mystery surrounding King Tut’s family and discover the identity of his father, we find that there are some archeologists who strongly suggest that this is most probably King Akhenaton,” said Dr Hawass.

Akhenaton was the first Pharaoh to advocate monotheism, not just in ancient Egypt, but in the world. Others believe that Akhenaton’s father, King Amenhotep III is a more likely candidate for Tutankhamen’s father.

As for King Tut’s mother, “If we follow the speculation mentioned above with regards to Tutankhamen’s father, his mother is most likely either Queen Tiye, the consort of King Amenhotep III or the extremely famous, Queen Nefertiti, the consort of King Akhenaton,” said Dr Hawass.

These questions are enigmatic, and archeologists are having a hard time trying to answer them.

According to Dr Hawass, “We have embarked upon the quest to solve the mysteries surrounding King Tut thanks to the two DNA analysis laboratories that we have access to, as well as the availability of a CT-Scan machine, through which we are able to know every single detail about a mummy.” (ANI)

Earliest stars in Universe may have been twins

Washington, July 10 (ANI): Astrophysicists, using extremely detailed computer simulations, have determined that the earliest stars in the universe formed not only as individuals, but sometimes also as twins.

The robust simulations of the early universe were created by astrophysicists Matthew Turk and Tom Abel of the Kavli Institute for Particle Astrophysics and Cosmology, and Brian O’Shea of Michigan State University.

“We used to think that these stars formed by themselves, but now we see from our computer simulations that sometimes they have siblings,” said Turk.

“These stars provide the seeds of next generation star formation, so by understanding them we can better understand how other stars and galaxies formed,” he added.

To make this discovery, the researchers created an extremely detailed computer simulation of early star formation.

Into this virtual universe, they sprinkled primordial gas and dark matter as it existed soon after the Big Bang, data they obtained from observations of the cosmic microwave background.

This mostly uniform radiation – a faint glow of radio waves spread across the entire sky – contains subtle variations that reflect the beginning of all structure in the universe.

The simulations focused on the first Population III stars: massive, hot stars thought to have formed a mere several hundred million years after the Big Bang.

As the researchers watched their simulated universe evolve, waves of gas and dark matter swirled through the hot, dense universe.

As the universe cooled, gravity began to draw the matter together into clumps. In areas rich with matter, stars began to form.

In one out of the researchers’ five simulations, a single cloud of dust and dark matter formed into “twin” stars: one with a mass equivalent to about 10 suns, and one with a mass equivalent to about 6.3 suns.

Both of them were still growing at the end of the calculation and will likely grow to many times that mass.

“We ran five of these calculations starting from the beginning of the universe, and to our surprise one of them was special,” said Abel.

“This opens a whole new realm of research possibilities. These stars could evolve into two black holes, which could have created gravitational waves we could detect with an instrument like the Laser Interferometer Gravitational Wave Observatory,” he added.

“This will help us fine-tune our models for how structure in the universe formed and evolved. Understanding the very early stars helps us understand what we see today,” Turk said. (ANI)

Nick Cannon has no grudges against Eminem

London, May 25 (ANI): Rapper Nick Cannon has admitted that he holds no grudges against Eminem, who made fun of his marriage to pop-diva Mariah Carey in a song.

Eminem a.k.a Marshall Mathers III, sparked a feud with Cannon after referring to his relationship with Carey in the track ‘Bagpipes From Baghdad’.

Eminem rapped in the song: “Nick Cannon better back the f**k up. I’m not playing, I want her back, you punk. Nick Cannon, you pr**k, I wish you luck with that f**kin’ whore.”

Cannon, who wed the Hero singer last year (08) after a whirlwind romance, was left fuming over the lyrics and vowed Eminem’s words will have “repercussions”.

Eminem later admitted that his lyrics were ‘a little harsh’, insisting he totally understood Cannon’s reaction.

Now, Cannon has also revealed that he has no grudges against the rapper.

“I never even considered it as beef. It’s not beef. It’s just, Hey, I heard the record. I said what I had to say. I feel exactly the same way by it and stick by everything, but I think it’s one of those things where it becomes that it’s not really that big of a deal, you know?” The Daily Star quoted Nick, as telling The Entertainment Weekly. Triple drug therapy shows promise in treating breast cancer

Washington, May 16 (ANI): Mayo researchers have suggested a triple drug therapy that may offer a promising new option for breast cancer.

They said that combining two chemotherapy drugs with trastuzumab (Herceptin) to treat women who have metastatic HER2+ breast cancer might offer physicians another choice in their treatment options.

“This is a very well tolerated regimen. The combination is a good example of an excellent therapeutic ratio: good activity and low toxicity,” said study’s senior investigator, Dr Edith Perez, director of Mayo Clinic’s Breast Center in Jacksonville.
Dr Winston Tan, a medical oncologist at Mayo Clinic said that the chemotherapy regimen was previously tested in Europe and demonstrated good anti-tumour activity and low toxicity, so Mayo researchers combined it with Herceptin.

They found that 67pct of the 45 patients responded to treatment, with their tumours decreasing in size by at least 30 percent.

“The results are encouraging, and would support a larger, randomized Phase III study,” he said.

“This is a Phase II study of this triple combination, so we would need to study this treatment against the standard best two-drug treatment in a randomized Phase III study to know if this triplet is more effective.

“This regimen seems to be a very reasonable choice, and it offers the added advantage that women who use it do not lose their hair,” he says. The drug combination used most commonly for patients with HER2+ breast cancer that has spread – paclitaxel or docetaxel with trastuzumab – always causes hair loss,” he added.

The study was presented at the 45th Annual Meeting of the American Society of Clinical Oncology (ASCO). (ANI)

Nick Cannon accuses Eminem of ‘racial bigotry’ after Carey rap

Washington, May 11 (ANI): American actor Nick Cannon has accused rapper Eminem, real name Marshall Mathers III, of “racial bigotry” after the latter made a derogatory new track mentioning Mariah Carey.

Cannon, 28, who married Carey, 39, last year, is furious with Eminem, 36, for putting his and his wife’s name in a controversial song, which is featured on his latest album ‘Relapse’.

In his latest LP, Eminem had recorded Bagpipes From Baghdad, and in it he comments on his alleged romance with Carey – one that she vehemently denies.

“Nick Cannon better back the f**k up. I’m not playing, I want her back, you punk. Nick Cannon, you pr**k, I wish you luck with that f**kin’ whore,” Contactmusic quoted Eminem as rapping in the track.

But Cannon isn’t going to let the star get away with his comments – vowing that Eminem’s words will have “repercussions”.

“A mediocre (at best) Eminem record that sounds like it was written in 2001.

At first, I thought it was old material that had been dug up from when dude ‘fantasised’ about having a pretend fling with Mariah. … But all of a sudden I hear my name in the verse! My first reaction was like, ‘This is his new s**t? Wow, that’s too bad’,” he wrote on his blog.

“Maybe I’m going too far, but I thought we got past the days where white men could spew vulgar obscenities at our beautiful queens and get away with it.

“What’s next? Are we going to let this trash say something horrible about our lovely first lady, Mrs. Michelle Obama? Or would Marshall have talked sideways out of his neck like this about Oprah Winfrey? This act of racist bigotry cannot go unnoticed.

“Calling my wife a ‘c**t’ and a ‘whore’ is way worse than anything Don Imus could have ever said. So trust, repercussions will be served. Anybody got Al Sharpton’s number?!

“So, Miss Marshall, I’m going to make you wish you never spoke my name and regret the ungodly things you said about my wife. This is going to be fun!

It’s however you want it! Just remember, you did this to yourself! Your legacy has now been tainted from this day forth! You will now be known as the rapper who lost to corny-ass Nick Cannon!” he added.

The blog, which featured on Cannon’s official website, has since been taken down due to an overload of comments.(ANI)

Personalized treatment for early lung cancer on the anvi

Washington, May 4 (ANI): Personalised treatments following surgery are now being offered for patients with early stages of lung cancer, experts at an international conference said.

The experts reckoned that cancer vaccines and targeted therapies could help patients with lung cancer after surgery.

“Personalizing therapy is the key strategy for longer and better survival in lung cancer,” said Prof Paris Kosmidis, head of the second Medical Oncology Department at Hygeia Hospital in Athens, Greece.

“This is particularly important for early stage disease when following surgery, decisions about preventive therapy are based on specific prognostic and predictive factors.”

Prof Walter Weder, head of thoracic surgery at the University of Zurich in Switzerland, adds: “At the meeting, several research groups will present data from new and ongoing studies that show how existing treatments might be combined with targeted therapies and new cancer vaccines. We hope that these new treatments will provide further progress.”

In one poster presentation at the meeting, researchers described the results of a study that sought to identify which patients are likely to benefit from an immune-boosting vaccine designed to help the immune system to recognize MAGE-A3, a protein that is expressed on about 30 percent of lung cancers.

Studies have shown that the treatment can help patients avoid or delay cancer recurrence after surgery.

At the conference, researchers will describe a gene signature that might predict which patients will benefit from the treatment.

“Because the benefits of vaccination will be limited to a subgroup of patients, strategies to define these patients by means of biomarkers such as a genetic signature are of major clinical relevance, as only these patients might be candidates for vaccination in the future,” said Weder.

A larger, phase III, trial of the vaccine is now underway.

The findings were put forward at the first European Multidisciplinary Conference in Thoracic Oncology (EMCTO) in Lugano, Switzerland. (ANI)

Diabetes, obesity, hypertension up mortality in hepatitis C patients

Washington, Apr 23 (ANI): Type 2 diabetes, obesity and hypertension are associated with overall and liver related mortality in hepatitis C (HCV) infected patients, a new research has claimed.

The specific impact of metabolic syndrome on mortality in hepatitis C patients has been revealed by new research to be presented on Sunday April 26 at EASL 2009, the Annual Meeting of the European Association for the Study of the Liver in Copenhagen, Denmark.

In HCV-infected patients, the top three predictors of liver related mortality were having higher body mass index (BMI), presence of insulin resistance (IR) and elevated serum cholesterol. Overall mortality in HCV patients was most linked to metabolic syndrome, higher BMI and hypertension.

Metabolic syndrome is a combination of medical problems that increase risks of cardiovascular disease and diabetes.

Recent data have suggested that metabolic syndrome is associated with adverse outcomes in HCV patients. This study set out to assess which aspects of metabolic syndrome are of most risk to such HCV patients and to quantify their specific impact on mortality.

Professor Zobair Younossi MD, MPH from the Center for Liver Diseases at Inova Fairfax Hospital and the Executive Director of Betty and Guy Beatty’s Center for Integrated Research, Virginia, USA, who led the study, said: “Exploring the risk factors associated with adverse outcomes in HCV patients helps us to better understand the complex nature of this highly prevalent disease.

“This study shows a clear association between key components of metabolic syndrome and mortality in HCV patients and demonstrates the importance of lifestyle improvements and coaching in the management of HCV patients, to potentially minimise the onset and impact of metabolic syndrome and its associated mortality risks.”

Researchers in this study utilised the Third National Health and Nutrition Examination Survey (NHANES III) and Linked Mortality Files. HCV was defined as positive HCV RNA by PCR assay. Subjects without other causes of chronic liver disease such as presumed NAFLD with elevated serum aminotransferases, excessive alcohol use, elevated transferrin saturation and positive hepatitis Bs antigen were designated controls without liver disease. HCV patients were compared to HCV-negative individuals and controls without liver disease using Rao-Scott chi-square statistics.

Adjusted hazard ratios for overall mortality and cause-specific mortality were calculated for HCV patients using persons without HCV. The Cox proportional hazard model was used for calculation of AHR for independent risk factors, and for the presence of HCV as a potential risk factor for overall mortality and cause-specific mortalities.

The cohort included 15,866 individuals with complete data. (ANI)
